Agilent Technologies 2100 bioanalyzer allows for semiquantitative detection of gene expression

PALO ALTO, Calif., Agilent Technologies Inc. announced a new application for the Agilent 2100 bioanalyzer in conjunction with the DNA 1000 LabChip( kit for the semiquantitation of mRNA expression.

The research on the quantitation of mRNA expression was conducted at the Forschungszentrum in Karlsruhe, Germanyand took advantage of comparative multiplex reverse transcription PCR. "Generating data with the Agilent 2100 bioanalyzer is one of the fastest and most robust methods for the analysis of quantitative or semiquantitative PCR applications I have experienced to date," said Dr. Eric Gottwald of the Forschungszentrum in Karlsruhe.

For this application, the primer dropping method, first described in 1994 by Wong et al, was employed. Until recently comparative PCR experiments depended on gel analysis, a slow, manual technique that does not provide accurate quantitation for multiple PCR. The DNA 1000 assay used with the Agilent 2100 bioanalyzer delivers accurate quantitation of multiple products over a wide range of concentration. Using this method semi-quantitative gene expression can be achieved, substituting or even replacing real-time PCR for certain applications. In contrast to real-time PCR, a multitude of genes can be analyzed in one single reaction.
