Sartobind® Q membrane adsorbers represent advanced chromatography technology designed for efficient protein capture and polishing applications. These void-volume-optimized systems feature specialized 4 mm and 8 mm bed heights engineered to deliver superior performance in both flow-through and bind and elute separations.
The 8 mm bed height configuration provides enhanced binding capacity with only 1.4 membrane volumes compared to 2.8 membrane volumes in 4 mm designs. This optimization enables sharp breakthrough curves and minimal elution volumes of approximately two bed volumes, eliminating back mixing of separated samples. Dynamic binding capacity reaches 22 g with bovine serum albumin at 2 g/L concentration.
Available in multiple formats from Sartobind® Nano 3 mL analytical units to Sartobind® Jumbo 5 L production scales, the product line ensures consistent scalability. Breakthrough behavior remains equivalent across all sizes when normalized per liter of membrane volume, with identical elution profiles maintaining reproducibility throughout scale-up processes.
The membrane adsorber technology incorporates flow channels optimized for minimal void volume, essential for achieving precise separation results. Sample flows across the membrane from upstream to downstream channels through a construction featuring either central cores in capsules or spacer elements in cassettes. This design enables complete purification cycles within 11 minutes at flow rates of 4 membrane volumes per minute.
Sartobind® Q strong ion exchangers excel in monoclonal antibody polishing applications, removing viruses, DNA, host cell proteins, leached Protein A, and endotoxins. The macro-porous structure with pore size greater than 3 µm features quaternary ammonium ligands covalently bound to the complete internal surface, resulting in high binding capacity combined with exceptional flow rates up to 30 bed volumes per minute.
For virus and virus-like particle applications, size exclusion effects are negligible, making these adsorbers suitable for purification of adeno- or lentiviruses with one order of magnitude higher dynamic binding capacity compared to conventional resins. The materials support more than 100 cycles of reuse without loss of binding capacity.
Flow-through polishing capabilities include virus clearance greater than 6 log, DNA removal below detection limits, host cell protein removal exceeding 99%, and endotoxin removal greater than 5 log while reducing column costs by 80%. High-resolution protein separation capabilities are demonstrated through analytical applications separating myoglobin, cytochrome C, and lysozyme mixtures at flow rates of 10 mL/min (3.3 membrane volumes/min).
