The Sartobind Convec® D represents a breakthrough in weak anion exchange membrane chromatography specifically engineered for downstream processing of lentiviral vectors and virus-like particles. This advanced chromatography solution leverages convective mass transfer mechanisms combined with optimized diethyl amino ligand chemistry to deliver exceptional infectious recovery rates exceeding 60% while maintaining gentle processing conditions essential for viral particle integrity.
The technology features stabilized reinforced cellulose membranes with precisely controlled 1.1 μm nominal pore size and optimized ligand density of 6-12 μmol/mL. This unique design eliminates size exclusion effects commonly encountered with traditional resin-based systems, making it particularly suitable for large biomolecules including lentiviruses and extracellular vesicles. The membrane architecture enables dynamic binding capacities of 2 × 10¹² VP/mL for clarified lentivirus cell culture supernatant at 10% breakthrough.
Linear scalability represents a key advantage, with formats spanning from Nano (3 mL) through production-scale capsules up to 1.2 L bed volume. Each format maintains consistent performance characteristics, facilitating seamless process transfer from development through commercial manufacturing. The capsules feature robust polypropylene construction with multiple connection options including Luer, sanitary clamp, and hose barb configurations to accommodate diverse processing requirements.
Gamma-irradiated versions are available with validated dose ranges of 25-50 kGy, supporting closed and aseptic bioprocessing workflows that reduce contamination and bioburden risks. The technology demonstrates excellent pH stability across 3-14 (short term) and 4-13 (long term), providing operational flexibility for various buffer systems and cleaning protocols.
