The inert (U)HPLC system 'Nexera XS inert' - experience newfound clarity!

Biomolecules such as phosphopeptides, nucleic acids or monoclonal antibodies tend to adsorb on metallic surfaces of UHPLC systems. The consequences: poor peak shape, insufficient sensitivity, distorted calibration curves and costly pre-passivation. The Nexera XS inert system solves these challenges by providing a completely metal-free sample flow path - with a simultaneous UHPLC pressure tolerance of up to 105 MPa (1050 bar).

Metal-free sample flow path for reliable bioanalysis

In the Nexera XS inert, special PEEK or ceramic materials are used in all areas with sample contact. The PLS capillary developed by Shimadzu enables operation at up to 105 MPa. Proprietary connection technology eliminates dead space volumes at the capillary connections, while a smart cap design allows tool-free column installation even at high pressures. The result is measurable: the Nexera XS inert, for example, achieves a four times improvement in the signal-to-noise ratio in phosphopeptide analysis compared to a standard UHPLC with stainless steel surfaces.

Online pH monitoring of the mobile phase with the pHM-40

When analyzing proteins or other biomolecules, the pH value of the mobile phase can strongly influence the chromatographic separation. The optional pHM-40 pH monitor adds an important control instrument to your Nexera XS inert. It continuously monitors the value of the mobile phase so that changes are detected in real time. For maximum data security, the pH value data is saved with the chromatogram and the calibration is stored in System Check Reports.

This system eliminates the need for prior passivation

In order to minimize the effects of adsorption, conventional HPLC systems are often passivated. For this purpose, samples with the target components are injected several times before the actual data acquisition. However, this procedure not only costs sample material and time, but also often makes it more difficult to achieve reliable results. The inert Nexera XS system eliminates the need for prior passivation: It provides reliable data from the first injection and throughout the entire analysis process.

Integration into digital analysis workflows

The Nexera XS inert system can be seamlessly integrated into the LabSolutions™ software platform. The additionally available LabSolutions™ MD software enables efficient development of analytical methods with an Analytical Quality by Design (AQbD) approach. All steps of the screening, optimization and validation phases of the method development process can be performed with LabSolutions™ MD - from the analysis of samples by automatically switching between the respective mobile phases and columns, to the use of analytical results to build a design space, to the selection of optimal analytical conditions and the evaluation of robustness (validation).
