DNA isolation from anti-coagulated whole blood for subsequent molecular analysis by PCR methods is well-established. In a standard laboratory setting, the blood is shipped in a blood collection vial containing the anticoagulant after phlebotomy. However, it might not always be possible to have blood samples shipped immediately, and for longer storage cooling is required. Freezing anticoagulated blood samples might interfere with the efficiency of DNA isolation afterwards. Thus, the collection and storage of whole blood samples on special filter paper has become more and more popular since it provides a simple and easy method to ship and to store the samples. Despite advances in transfection reagents and methods, there are still a number of cell lines which are difficult to transfect, such as T47D mammary carcinoma cells. Using a ß-galactosidase reporter plasmid, the transfection of T47D cells with FuGENE® HD Transfection Reagent has been optimized. Several scientific studies support the claim that natural scrapie is associated with polymorphisms at three codons within the sheep prion protein (PrP) gene. Genotyping of these codons (codon 136 [alanine, valine], 154 [arginine, histidine], and 171 [glutamine, arginine, histidine]) might enable breeding of sheep flocks with high resistance to scrapie. Fast and reliable high-throughput genotyping of sheep is a major issue, since scrapie is a serious problem in several European countries.
