Roche’s xCELLigence System RTCA HT Instrument performs real-time impedance-based label-free measurement of cell behavior for high throughput applications. It´s technology is compatible with workflows carrying out automated screening using the 384-well culture plate format.
One to four specialized E-Plates 384 with microelectrodes at the bottom of each well can be controlled independently, using up to four RTCA HT stations, driven by dedicated software. Measurements in each 384-well plate are obtained in parallel. The RTCA HT Instrument, comprising an analyzer, up to four stations with E-Plates 384, and the control unit, is easily integrated into automated 384-well plate and liquid handling systems.
The RTCA HT Instrument was used for GPCR and cytotoxicity assays by Prof. Matt Cooper’s research team in the Institute of Molecular Bioscience, University of Queensland, Brisbane, Australia. Prof. Cooper is a pioneer of label-free technologies with two books on the subject: “Label-Free Technologies for Drug Discovery (Wiley-Blackwell) and “Label-free Biosensors: Techniques and Applications” (Cambridge University Press).
Prof. Cooper reported, “We see the advent of 384-well screening systems based technologies, such as electrical impedance, as highly significant for GPCR screening of ligand-directed signaling. Label-free can really challenge current screening paradigms, as the readout is non-invasive, temporal, cumulative, and most importantly, signaling-pathway independent.”
The xCELLigence System measures cellular electrical impedance producing a label-free, non-invasive and kinetic readout of cellular status. Impedance recording is ideal for many different types of experiments investigating how cells respond in vitro when challenged with compounds and other stimuli. Data produced by the system are incisive and information-rich, with the potential of being more biologically meaningful than data produced by more invasive platforms using exogenous labeling.
