  • Study uncovers how herpesvirus spreads

Herpesviruses are thrifty reproducers - they only send off their most infectious progeny to invade new cells. Two Cornell virologists recently have discovered how these viruses determine which progeny to release. The College of Veterinary Medicine researchers report in Proceedings of the National Aca more

  • Deciphering how CD4 T cells die during HIV infection

Scientists at Gladstone Institute of Virology and Immunology have solved a long-standing mystery about HIV infection–namely how HIV promotes the death of CD4 T cells. It is the loss of this critical subset of immune cells that leads to the development of AIDS. Most immune cells that die during HIV i more

  • Real-Time Multiplex PCR of five Different DNA Targets Using the LightCycler® 480 System

One of the most interesting aspects of real-time PCR based on detection of fluorophoric-labeled oligonucleotides, such as Hydrolysis probes, and Molecular Beacons, is the possibility of being able to detect conveniently multiple targets in the same PCR reaction (multiplex PCR). Ideally, a real-time multiplex PCR should be able to detect, differentiate, and provide a quantitative result for many different targets without a single target influencing the detection of one of the others (cross-talk) and without loss of sensitivity. It is evident that due to the limited number of fluorophoric labels available and the significant overlap in their emission spectra, quantification of multiplex reaction products is difficult and often not possible for more than two targets. more

  • Fugene® HD Transfection Reagent: Superior Performance for Challenging Expression Studies

The ability to efficiently introduce foreign molecules into cells constitutes an important tool in any laboratory and facilitates studies of gene regulation and protein function. A plethora of transfection methods have been developed for transfer of DNA into mammalian and animal cells. Those methods either utilize the physicochemical properties of the vector-carrier of the DNA or use mammalian viruses. In the first group of methods, some classic techniques are well-known, such as: calcium phosphate, DEAE-dextran, electroporation liposome-mediated delivery, direct microinjection/biolistic particle delivery, and non-liposomal multi-component reagents. Viral vector systems use both DNA (Adeno-) and RNA (Retro-, Lenti-) viruses. more

  • Efficient Transfection of VeroE6 Cells with DNA and RNA for Luciferase Reporter Assays Using FuGENE® HD Transfection Reagent

The interferon (IFN) system of mammalian cells is the first line of defense against viral infections. Following infection, danger signals such as double-stranded RNA or single-stranded RNA with an unmodified 5´ triphosphate end (e.g., VSV-RNA) are recognized by membrane-bound Toll-like receptors and/or cytoplasmic receptors (MDA5, RIG-I). This initiates a signal cascade which leads to the production of type I interferons. By autocrine and paracrine stimulation via the IFN-a/b receptor the expression of antiviral proteins is induced in the infected cell as well as in the surrounding cells. more
