TOYOPEARL AF-rProtein A-650F combines a high binding capacity at short residence time and fast mass transfer kinetics with alkaline stability. Protein A based affinity chromatography is the industry standard for the capture of monoclonal antibodies (MAbs) in downstream processing.
Rising fermentation titres have triggered the demand for resins that increase downstream capacity and efficiency. TOYOPEARL AF-rProtein was designed to meet these requirements. The typical dynamic binding capacity for IgG is higher than 30 mg/mL resin at two minutes residence time. Thus, TOYOPEARL AF-rProtein A offers the opportunity to improve process economics by increasing both, throughput and robustness.
The high binding capacity helps to minimize the resin and buffer volumes needed for a given target load. Applying higher flow rates reduces loading and cycle times in lab scale and process scale purifications.
A newly developed recombinant ligand (non-animal-derived) has been engineered to maintain capacity even after repeated exposure to alkaline solution. Its multipoint attachment to the TOYOPEARL polymeric matrix further enhances chemical and thermal stability.
In practice this pays off for a low level of protein A leaching and also for a high resistance to alkaline solutions employed in cleaning-in-place (CIP) procedures. As a consequence the number of cycles can be increased when compared to Protein A resins with lower caustic stability. Due to its rigid polymer matrix the resin also provides excellent pressure flow characteristics. It is therefore ideally suited for industrial scale purification of antibodies in large process columns.
TOYOPEARL AF-rProtein A belongs to a whole series of chromatography media that were optimized for antibody purification: TOYOPEAL GigaCap ion exchanger, HIC-600 resins for hydrophobic interaction and the hydrophobic cation exchanger TOYOPEARL MX-Trp.
For fast screening of resins and method parameters we offer pre-packed ToyoScreen cartridges with 1 mL and 5 mL bed volume or ToyoScreen RoboColumns with 0.2 or 0.6 mL volume for high throughput screening on robotic workstations.
