  • Real-Time PCR Quantification of Plant miRNAs Using Universal ProbeLibrary Technology

    MicroRNAs (miRNAs) are naturally occurring, highly conserved families of transcripts (18-25 nt in length) that are processed from larger hairpin precursors. Both plant and animal genomes encode miRNAs, and to date, there are over 4000 mature miRNA transcripts annotated. Although miRNAs represent a relatively abundant class of transcripts, their expression levels vary greatly among species and tissues. Less abundant miRNAs routinely fail detection using technologies such as cloning, northern hybridization and microarray analysis. Here, we present a real-time quantification method for accurate, sensitive, and economical detection of plant miRNAs based upon the stem-loop RT primer approach using an miRNA-specific primer coupled with a stem-loop specific Universal ProbeLibrary (UPL) probe. This approach represents a highly specific, sensitive, and easy-to-apply system to detect miRNAs. more

  • Microsatellite Analysis of Grapevine Varieties by High-Resolution Amplicon Melting Analysis

    Certification of grapevine scion and rootstock varieties is becoming an increasingly important issue in viticulture. Many rootstocks are derived from the same parentage - for example 5C, SO4, 5BB, 125AA, and 420A Mgt are all used in New Zealand and are all derived from crosses between Vitis berlandieri and Vitis riparia. This use of limited genetic stock is not uncommon when compared with other countries and means that cultivar determination based on ampelography (visual characteristics of grapevines) can be difficult at times due to the high similarities. more

  • Transfection of T47D Mammary Carcinoma Cells Using FuGENE HD® Transfection Reagent

    Despite advances in transfection reagents and methods, there are still a number of cell lines which are difficult to transfect, such as T47D mammary carcinoma cells. Using a ß-galactosidase reporter plasmid, the transfection of T47D cells with FuGENE® HD Transfection Reagent has been optimized. Using a variety of FuGENE® HD Transfection Reagent-to-DNA ratios, we have shown through RT-PCR and ß-galactosidase enzyme activity assays that FuGENE® HD Transfection Reagent is able to transfect this cell line with high efficiency. more
