    Exocrine glands have been shown to be potential sources of pluri-/multipotent adult stem cells that are highly versatile as demonstrated for pancreas, testis, salivary glands, and sweat glands. Primary cultures of these adult stem cells have the capacity for extended self-renewal and are able to differentiate in vitro into cell types of the three embryonic germ layers. The Fraunhofer EMB established a standardized isolation procedure that was successfully applied to the isolation of proliferating cells from different animal tissues and human biopsies. With this technique, tissue specimens are ­generally dissociated in a two-step protocol using collagenase. The gland-derived cell populations adhere to the culture dish and start to proliferate. During in vitro propagation, these glandular stem cells exhibit a population doubling of at least 30, depending on the organ and the species more

    For the in vitro testing of pharmaceutical, cosmetic and chemical products conventional cell cultures are increasingly being replaced by three-dimensional (3D) cell culture models with organ-specific characteristics. Using tissue engineering techniques, a living human skin equivalent has been developed at the Fraunhofer IGB which mimics the normal human skin. The fabrication of the 3D skin model requires the isolation and growth of primary keratinocytes and fibroblasts from human skin tissue. For the isolation of the cells it is necessary to dissociate the skin tissue in the dermal and epidermal layer without destroying the cells. Usually, cell isolation is performed in a two-step protocol. Following dissociation of both layers with dispase, collagenases are used to release the fibroblasts from the dermis. The goal of all cell isolation procedures is to maximize the yield of dissociated cells that are viable and functionally active. more
