Sennae fructus

Analysis and evaluation of the new draft monographs for the European Pharmacopoeia

The analytical methods of the monographs in the European Pharmacopoeia of Sennae fructus are becoming more important because of increasing cultivating areas. An analytical differentiation between the plant sources of the commercial drug “Sennae fructus”, Cassia acutifolia and Cassia angustifolia, is currently impossible by means of the methods of the monographs of the European Pharmacopoeia. Concerning the content of hydroxyanthracene derivatives of these drugs, members of the expert group 13 A of the European Pharmacopoeia Commission suggested replacing the photometric assay by an HPLC method. Therefore, in this study HPLC-UV(DAD)-ESI/MS and HPLC-ESI/MS/MS measurements were performed to evaluate the efficiency of this proposed HPLC method, which allowed the separation and assignment of 32 constituents of a Sennae fructus extract: 19 hydroxyanthracenes, 8 flavonoids, 2 benzophenones, and 3 naphthalenes. Moreover, this HPLC analysis enabled a differentiation between extracts of Cassia angustifolia und Cassia acutifolia by means of tinnevellin, tinnevellin-8-glucosid, 6-hydroxymusicin-glucoside and an unknown derivative.
